Over Optimization basically adheres to the principal, "If some is good, more is better". The most often cited example is "keyword stuffing", which means using as many keywords as you can as much as possible - Title Tag, meta tags, and the content of the website. For off-page optimization, it means using links from too many of the same site, or same type of site. For example, using a large quantity of .edu links because they are worth more to the search engines.
How does this effect your website? Ironically, if you design your website for the search engines, the search engines will not like your website. As a result, your website will either not rise in the SERPs, or will lose whatever ranking it has.
