How can we check either the links are no follow or do follow ?
If you want to find the links is either do follow or no follow then you have to download online tools. Even some are Firefox add-ons and also chrome extensions assisting you to highlight do follow or no follow. These are the tools

SEOmoz toobar is best to check the link is do follow or no follow,
For viewing it manually you can view its page source. The links are with or with do follow tags are considered as Do Follow where as the links with clear no follow are considered as no follow.
As adrian said plus you can use browser's "inspect element" with a right-button click on the actual link.
you should installed seo quake at your browser then you can check no follow or do follow links easily
Do Follow : When Google bots are crawling a website they look for external links point to other websites. If this links is “do follow” then search engines will follows the link and so link juice gets passed.
No Follow : If a search engine finds a “no follow” backlink it will just ignore the link and will not get deeper into crawling and no link juice is passed.
I think in the seo process both no follow and do follow links matter
do follow count as back link to the site from search engine. It's important for site ranking to get quality back links.no follow doesn't count as back link to the site from search engine but it can be helpful as an traffic generate source if it's come from high pr sites.
It's very simple, the NoFollow extension outlines nofollow links which are marked with rel='nofollow' and links on pges with nofollow robots meta tag. It outlines it as soon as the webpage gets loaded completely.its basically a dofollow and nofollow checker.

Install nofollow adon/extension to your blog gives you the detail about each link in the page and highlight if the link is nofollow or dofollow.
Use Greasemonkey add-on for Mozilla Firefox. it is simple and it does not install any third party tools which cone with the seomoz toolbar .
To check the nofollow sites
To check manually go to the page source and search the word "no follow", If the page contains the word no follow, then the site is nofollow, If it doesn't contains the nofollow tag, then the site is do follow. Otherwise you can check the no follow sites using some add-ons. Ex: for mozilla, you can use "Greasemonkey", for chrome, you can use "No follow" plugin, and SEOQuack also is the best tool to check nofollow sites.
Nofollow links are identified as <a href="sign.php" rel="nofollow">sign in</a> in the coding of the web page.
