How much budget would you allocate for a startup business like selling online items (ie. through affiliates) ? I mean how much do you pay for directory submission, online advertising, newspaper advertising and social bookmarking for your websites? Would $250 be enough?

Hard question to answer, it depends on how much work you do yourself or outsource, off line newspaper ads can be costly, unless you are thinking small shopper/penny saver type papers, and for online ads. That depends on if you are in a high paying PPC niche, if you plan on doing PPC.

You would need to nail down your marketing plan so you would know.

The budget you need depends completely on the type of business that you set up. You can start a free blog on something like blogger, without spending any money. At the other end of the spectrum, if you want to buy Yahoo and compete directly with Google you will need a phenomenal budget.

You should plan your online or offline business in detail and be careful to include all costs in your projections so that you can get an accurate estimate of the total budget you need to establish the business.
